Morpholinos屬于第三代反義寡核苷酸,由Gene Tools公司獨家研制、開發和提供合成服務。
Morpholinos主要通過阻斷蛋白質翻譯過程而達到抑制目標基因功能的作用,目前已經廣泛用于發育生物學和細胞學實驗中,并取得了非常好的實驗效果。
Morpholinos以嗎啉環類似物為基本骨架,這種獨特的結構特性使得Morpholinos具有以下幾個特點:
n 與其他類型的反義寡核苷酸相比,Morpholinos能更好地抵抗核酸酶的作用
n Morpholinos與靶序列結合后通過空間位阻效應發揮作用,不激活 RNaseH,不引起目標基因mRNA 的降解
n 針對翻譯起始位點區設計的Morpholinos用于抑制蛋白質的翻譯,針對pre-mRNA剪切位點區設計的Morpholinos可影響mRNA剪切,得到不同的轉錄本,可以方便地用于目標基因結構域的突變研究
n 與靶序列結合能力強,可以滲入mRNA的二級結構,并且特異性好
配合Morpholinos的使用,Gene Tools公司開發了相應的輸送系統,可以安全、高效地將Morpholinos 輸送至胞漿內,因此,Morpholinos 也可以用于遺傳學研究和新藥靶標的發現以及驗證。此外,Morpholinos可以標上熒光基團、親和標簽以及活性反應基團,以供不同的研究需要。
